I think what helps is that I went through a phase where I was pretty insecure about what kind of idea I was even making. I really wanted it to be one with mass appeal and that could have a good fanbase. Even asking myself before I would start if it was attached to anything popular. Then using blood, crystals, and other magically reactive regents you can then use these rune circles to apply various effects to the surrounding area or enchant objects. With the more time you give to a rune circle to settle, and the deeper and more runes you etch in basically over time it gets stronger to the point of physically and spiritually warping the region around you with the effects you want.

With these different effects being like inspiring growth which will cause plants to go wild and people to automatically heal inside, or summoning darkness which will create never ending hallways and traps all throughout the house. Basically the aspect you choose will have negative and positive attributes it'll apply and a big part about setting these runes up is balancing those effects so they don't go crazy.

You can definitely apply runes to people, but they have to be runes representing spirits rather than concepts. Raw, natural concepts like darkness and growth will horribly mutate you since you're basically creating a pact or portal between your body and a realm of pure cancerous growth or infinite darkness. You have to find a demon, angel, god, or even ghost to form living runes.

Since these spirits can regulate the energy they're giving, you can create pacts that can limit and control the influence they can have on your bodies. Of course the downside of this is unlike the concepts which demand nothing for using their power since their only goal is to expand, these spirits can make rules for you to follow. Which will depend on their personalities and goals.

Thankfully they have Charlie.

Which neatly leads to Charlie. She's the anti-christ full stop. And not just that but she's an outsider as well. Someone from a different plane and thus not forced into the rules and roles of this one. So in broader spiritual terms she represents change, neither good or bad. But the more power she has, the more her magic and their magic combine and forcibly changes this world to more fit hers.

So in some sense she has unlimited power, since her soul is directly tied to hell which is more of a concept and thus practically infinite. But in the other sense, she's pretty limited in how much of that she can physically use. Not just personality wise, but her magic and this world's magic don't mix very well. So if she gives you her rune, you can't use anybody else's. You are directly tied to her.

If you die, you don't go to any other heaven or hell or any realm owned by the various spirits. You go to Charlie's realms. You are directly sucked into her soul and exists in this separate plain she is the god of. That's why she's very careful when she gifts it because to her she's damning you to her personal hell.
